Indication
Treatment of pulmonary arterial hypertension (PAH).
Dosage (summary)
Start at 5 mg once daily, may increase to 10 mg if tolerated.
Onset of Action / Duration
Onset: 2 hours, Duration: 13.6 to 17.9 hours.
Special Populations
- Elderly
- Hepatic impairment
- Renal impairment
Pregnancy & Breastfeeding
Contraindicated in pregnancy and lactation; may cause serious birth defects.
Key Drug Interactions
- Ciclosporin A
- Rifampin
- Ketoconazole
Contraindications
- Hypersensitivity to ambrisentan
- Severe hepatic impairment
- Idiopathic pulmonary fibrosis
Common side effects
- Anaemia
- Headache
- Palpitations
- Flushing
- Fluid retention
Counselling Points
- Monitor liver function regularly
- Report signs of liver injury
- Use reliable contraception during treatment
Serious warnings
- Hepatic injury
- Auto-immune hepatitis
- Fluid retention leading to heart failure
