Indication

Treatment of pulmonary arterial hypertension (PAH).

Dosage (summary)

5 mg once daily, may increase to 10 mg if tolerated; 10 mg once daily with tadalafil.

Special Populations

  • Elderly
  • Renal impairment
  • Hepatic impairment

Pregnancy & Breastfeeding

Contraindicated in pregnancy and lactation; potential teratogenic effects.

Key Drug Interactions

  • Ciclosporin A
  • Ketoconazole
  • Rifampicin
  • Sildenafil
  • Warfarin

Contraindications

  • Hypersensitivity to ambrisentan
  • Pregnancy
  • Severe hepatic impairment
  • Idiopathic pulmonary fibrosis

Common side effects

  • Anaemia
  • Headache
  • Palpitations
  • Flushing
  • Fluid retention

Counselling Points

  • Monitor for signs of hepatic injury
  • Avoid in pregnancy
  • Report any signs of anaemia

Serious warnings

  • Hepatic injury
  • Auto-immune hepatitis
  • Potential for hypotension
